Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000003.11:g.(?_66119284)_(66429348_?)dup, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the duplication of exons 1-11 (i.e. the full coding sequence) of the SLC25A26 gene. A presumed nomenclature of c.(?_-729)_(*1117_?)dup has been designated for the purposes of this classification. It has been assumed that this is a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). Since exact breakpoints of this duplication are not known, it might extend beyond the assayed region of the SLC25A26 gene, including other flanking genes. A large duplication variant (1168229 bp) involving the SLC25A26 was found at a frequency of 4.6e-05 in 21694 control chromosomes (gnomAD, Structural Variant Dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.(?_-729)_(*1117_?)dup in individuals affected with Combined Oxidative Phosphorylation Deficiency 28 and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
