Uncertain significance for Hypokalemic periodic paralysis, type 1 — the classification assigned by Johns Hopkins Genomics, Johns Hopkins University to NM_000069.3(CACNA1S):c.2553G>A (p.Met851Ile), citing ACMG Guidelines, 2015: This CACNA1S missense variant is absent from a large population dataset, and has not been reported in ClinVar nor the literature, to our knowledge. Of two bioinformatics tools queried, one predicts that the substitution would be damaging, while the other predicts that it would be tolerated, and the methionine residue at this position is evolutionarily conserved across all except one of the species assessed. We consider the clinical significance of c.2553G>A; p.Met851Ile in CACNA1S to be uncertain at this time.
