This variant lies in the ABCD1 gene (transcript NM_000033.4) at coding-DNA position 1747, where G is replaced by C; at the protein level this means replaces valine at residue 583 with leucine — a missense variant. Submitter rationale: Variant summary: ABCD1 c.1747G>C (p.Val583Leu) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 153962 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1747G>C has been observed in a female via newborn screening for adrenoleukodystrophy (example: Matteson_2021). This report does not provide unequivocal conclusions about association of the variant with disease. A different variant affecting the same codon has been classified as pathogenic by our lab (c.1747G>A, p.Val583Met), supporting the critical relevance of codon 583 to ABCD1 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
