NM_001231.5(CASQ1):c.984+1G>A was classified as Likely pathogenic for Myopathy due to calsequestrin and SERCA1 protein overload by Diagnostics Services (NGS), CSIR - Centre For Cellular And Molecular Biology, citing ACMG Guidelines, 2015: The c.984+1G>A variant is not present in 1000 Genomes, EVS, ExAC, gnomAD, Indian Exome Database or our in-house exome database. This variant has neither been published in literature with CASQ1-related conditions nor reported to ClinVar, HGMD or OMIM databases in any affected individuals. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant can disrupt the consensus splice site. In-silico pathogenicity prediction programs like MutationTaster2, CADD, Varsome, Franklin etc predicted this variant to be likely deleterious however these predictions were not confirmed by published functional/translational studies.
