Pathogenic for Deficiency of UDPglucose-hexose-1-phosphate uridylyltransferase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000155.4(GALT):c.382G>A (p.Val128Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 128 of the GALT protein (p.Val128Ile). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with galactosemia (PMID: 22944367, 31954591). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GALT protein function. For these reasons, this variant has been classified as Pathogenic.
