Submitter rationale: ABCB11 p.Lys461Glu (c.1381A>G) is a missense variant that changes the amino acid at residue 461 from Lysine to Glutamic acid. This variant has been observed in at least one proband with an ABCB11-related disorder (PMID:37361697;21822150;9806540). It has been observed in trans with a pathogenic or likely pathogenic variant (PMID:21822150). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:17947449).
