This variant lies in the USH2A gene (transcript NM_206933.4) at coding-DNA position 7234, where G is replaced by A; at the protein level this means replaces valine at residue 2412 with methionine — a missense variant. Submitter rationale: Variant summary: USH2A c.7234G>A (p.Val2412Met) results in a conservative amino acid change located in the fibronectin type III domain (IPR003961) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 250980 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7234G>A has been reported in the literature in a setting of multigene panel testing in at least one individual affected with retinitis pigmentosa (e.g. Maeda_2018, Inaba_2020). These reports do not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
