This variant lies in the VWF gene (transcript NM_000552.5) at the canonical splice donor site of the intron immediately after coding-DNA position 3538, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: VWF c.3538+1G>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 5' splicing donor site, and one predicts the variant weakens a cryptic 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 53836 control chromosomes. c.3538+1G>C has been reported in the literature in at least one individual affected with type 2M Von Willebrand Disease (e.g. Shen_2016).
