A presumed nomenclature of c.(1331+1_1332-1)_(1482+1_1483-1)dup has been designated for the purposes of this classification. It is assumed to be a tandem duplication in direct orientation (Richardson_GIM_2018, Newman_AJHG_2015). This Copy Number Variant (CNV) is expected to alter the reading frame and predicted to result in a truncation (p.Val495Phefs*3 in DOVE database) or absence of the encoded protein due to nonsense mediated decay (NMD). The variant was absent in 14523 control chromosomes (gnomAD, Structural Variants dataset). c.(1331+1_1332-1)_(1482+1_1483-1)dup has been reported in the literature in multiple individuals affected with Duchenne and Becker muscular dystrophies (White_2006, White_2002, van Spaendonck-Zwarts_2012, Ling_2020, Alghamdi_2021, Nallamilli_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One submitter has cited clinical-significance assessments for this variant to ClinVar after 2014 and has classified the variant as pathogenic.
