NM_000518.5(HBB):c.193G>A (p.Gly65Ser) was classified as Uncertain significance by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: The beta-globin (HBB) c.193G>A (p.Gly65Ser) variant (also known as Hb Hezhou) has not been described in online databases. It also has not been reported in large, multi-ethnic general populations (http://gnomad.broadinstitute.org). In the published literature, the variant has been reported in two heterozygous Chinese individuals that were clinically normal and the protein was stable (PMID: 33843396 (2021)), however further research is needed. The variant has also been reported as part of a complex allele with c.400G>C (p.Val134Leu) known as Hb Extremadura that does show instability (PMID: 21797709 (2011)). Analysis of the Hb Hezhou variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
