Pathogenic for Birt-Hogg-Dube syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_144997.7(FLCN):c.1367_1398del (p.Asp456fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This premature translational stop signal has been observed in individual(s) with Birt-Hogg-Dubé syndrome (PMID: 25519458). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asp456Glyfs*19) in the FLCN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in FLCN are known to be pathogenic (PMID: 15852235).
