This variant has been observed in at least one proband with features of ATP8B1-deficiency (PMID:29973134;24342334;21342337). It has been observed in trans with a pathogenic or likely pathogenic variant (PMID:21342337). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:29104077). At least one splicing study demonstrated no effect on splicing (PMID:33437900). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ATP8B1 p.Glu981Lys (c.2941G>A) as a likely pathogenic variant.
