Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.526T>C (p.Tyr176His), citing Ambry Variant Classification Scheme 2023: The p.Y176H variant (also known as c.526T>C), located in coding exon 5 of the TSC1 gene, results from a T to C substitution at nucleotide position 526. The tyrosine at codon 176 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
