Pathogenic for Citrin deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014251.3(SLC25A13):c.848+1del, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gly283Aspfs*3) in the SLC25A13 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SLC25A13 are known to be pathogenic (PMID: 10369257, 14680984, 27405544). This variant is present in population databases (rs758135569, gnomAD 0.01%). This premature translational stop signal has been observed in individual(s) with citrin deficiency (PMID: 27405544). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as c.845_c.848+1delG; c.848+1del. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
