This variant lies in the BRCA2 gene (transcript NM_000059.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 9118, where A is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 23 of the BRCA2 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with breast cancer and/or ovarian cancer (PMID: 9150152, 30103829). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 267718). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 22632462; Invitae).
