Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_024120.5(NDUFAF5):c.946-2A>C, citing LabCorp Variant Classification Summary - May 2015: Variant summary: NDUFAF5 c.946-2A>C is located in a canonical splice-site in the last intron and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing and loss of NDUFAF5 function. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a 3' acceptor site. Three predict the variant creates a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.6e-05 in 250512 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.946-2A>C in individuals affected with NDUFAF5-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
