Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000059.4(BRCA2):c.7007+2T>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the BRCA2 gene (transcript NM_000059.4) at the canonical splice donor site of the intron immediately after coding-DNA position 7007, where T is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: ClinVar contains an entry for this variant (Variation ID: 267669). For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site results in skipping of exon 13 and/or exon 12 and introduces a premature termination codon (PMID: 21394826, 22505045). The resulting mRNA is expected to undergo nonsense-mediated decay. Based on a multifactorial likelihood algorithm using genetic, in silico, and/or statistical data, this variant has been determined to have a high probability of being pathogenic (PMID: 21990134). Disruption of this splice site has been observed in individual(s) with breast and/or ovarian cancer (PMID: 29446198). This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 13 of the BRCA2 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product.
