This variant lies in the MOCS1 gene (transcript NM_001358530.2) at coding-DNA position 949, where C is replaced by T; at the protein level this means replaces arginine at residue 317 with cysteine — a missense variant. Submitter rationale: Variant summary: MOCS1 c.949C>T (p.Arg317Cys) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4e-06 in 251402 control chromosomes. c.949C>T has been observed in multiple individuals affected with Molybdenum Cofactor Deficiency (e.g. Lubout_2018, Yoshimura_2019). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
