Likely pathogenic for Bifunctional peroxisomal enzyme deficiency — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000414.4(HSD17B4):c.350A>T (p.Asp117Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HSD17B4 c.350A>T (p.Asp117Val) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 250326 control chromosomes (gnomAD). c.350A>T has been observed in individuals affected with clinical features of D-Bifunctional Protein Deficiency (Bae_2020, Yamamoto_2021, Ko_2023). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
