The variant was absent in 250182 control chromosomes. c.394G>A has been reported in the literature in trans with a pathogenic variant in a compound heterozygous individual affected with Maple Syrup Urine Disease (Rodriguez-Pombo_2006). BCKD activity measured in intact fibroblasts from this individual was approximately 1% of normal activity, suggesting the variant disrupts protein function (Rodriguez-Pombo_2006).
