Pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024675.4(PALB2):c.3201+1G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PALB2 gene (transcript NM_024675.4) at the canonical splice donor site of the intron immediately after coding-DNA position 3201, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 11 of the PALB2 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with clinical features of PALB2-related conditions (PMID: 22241545, 25099575, 28888541, 34846068). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 34846068; Invitae). For these reasons, this variant has been classified as Pathogenic.
