Likely benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_022162.2(NOD2):c.241C>G (p.Leu81Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: NOD2 c.241C>G (p.Leu81Val) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 6.8e-05 in 251066 control chromosomes. The observed variant frequency exceeds the estimated maximal expected allele frequency for disease-causing variants in NOD2. c.241C>G has been observed in individual(s) affected with clinical features of Blau syndrome (Martin_2003). These report(s) do not provide unequivocal conclusions about association of the variant with Blau syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 14597055). ClinVar contains an entry for this variant (Variation ID: 267320). Based on the evidence outlined above, the variant was classified as likely benign.
