Uncertain significance for Lymphedema-posterior choanal atresia syndrome — the classification assigned by Clinical Genomics Laboratory, Washington University in St. Louis to NM_005401.5(PTPN14):c.1574C>T (p.Pro525Leu), citing ACMG Guidelines, 2015: The PTPN14 c.1574C>T (p.Pro525Leu) variant was identified at a near heterozygous allelic fraction. This variant, to our knowledge, has not been reported in the medical literature. This variant has been reported in one case in the cancer database COSMIC (Genomic Mutation ID: COSV652). This variant is only observed on 2/152174 alleles in the general population (gnomAD v.2.1.1), indicating it is not a common variant. Computational predictors suggest that the variant does not impact PTPN14 function. Due to limited information and based on ACMG/AMP guidelines for variant interpretation (Richards S et al., PMID: 25741868), the clinical significance of this variant is uncertain at this time.
