Uncertain significance for Pityriasis rubra pilaris; Psoriasis 2 — the classification assigned by Clinical Genomics Laboratory, Washington University in St. Louis to NM_001366385.1(CARD14):c.251G>A (p.Gly84Glu), citing ACMG Guidelines, 2015: The CARD14 c.251G>A (p.Gly84Glu) variant, to our knowledge, has not been reported in the medical literature and is only observed on 2/282,740 alleles in the general population (gnomAD v.2.1.1), indicating it is not a common variant. Computational predictors are uncertain as to the impact of this variant on CARD14 function. Due to limited information, and based on ACMG/AMP guidelines for variant interpretation (Richards S e t al., PMID: 25741868), the clinical significance of this variant is uncertain at this time.
