Uncertain significance for Hennekam lymphangiectasia-lymphedema syndrome 1 — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_133459.4(CCBE1):c.140G>C (p.Cys47Ser), citing ACMG Guidelines, 2015: The missense variant c.140G>C(p.Cys47Ser) in CCBE1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. The amino acid Cysteine at position 47 is changed to a Serine changing protein sequence and it might alter its composition and physico-chemical properties. The variant is predicted to be damaging by SIFT. The amino acid change p.Cys47Ser in CCBE1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
