Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.8517C>A (p.Tyr2839Ter), citing Ambry Variant Classification Scheme 2023: The p.Y2839* pathogenic mutation (also known as c.8517C>A), located in coding exon 19 of the BRCA2 gene, results from a C to A substitution at nucleotide position 8517. This changes the amino acid from a tyrosine to a stop codon within coding exon 19. This alteration has been identified in multiple individuals diagnosed with breast cancer (Yang X et al. PLoS One, 2015 Apr;10:e0125571; Sun J et al. Clin Cancer Res, 2017 Oct;23:6113-6119; Li JY et al. Int J Cancer, 2019 01;144:281-289). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
