Submitter rationale: The c.2960dupA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a duplication of A at nucleotide position 2960, causing a translational frameshift with a predicted alternate stop codon (p.N987Kfs*2). This mutation was detected in an Ashkenazi Jewish Israeli breast cancer patient diagnosed at age 41 (Barnes-Kedar I et al. Breast Cancer Res Treat, 2018 Nov;172:151-157). This alteration was also identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum Mutat, 2018 05;39:593-620). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
