NM_007294.4(BRCA1):c.5155dup was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 5155, duplicating one base. Submitter rationale: The c.5155dupG pathogenic mutation, located in coding exon 17 of the BRCA1 gene, results from a duplication of G at nucleotide position 5155, causing a translational frameshift with a predicted alternate stop codon (p.V1719Gfs*6). This alteration was identified in a large, worldwide study of BRCA1/2 mutation positive families (Rebbeck TR et al. Hum. Mutat. 2018 05;39:593-620). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
