Uncertain significance for Loeys-Dietz syndrome 4 — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_003238.6(TGFB2):c.959G>A (p.Arg320His), citing ACMG Guidelines, 2015. This variant lies in the TGFB2 gene (transcript NM_003238.6) at coding-DNA position 959, where G is replaced by A; at the protein level this means replaces arginine at residue 320 with histidine — a missense variant. Submitter rationale: The missense c.959G>A(p.Arg320His) variant in TGFB2 gene has not been reported previously as a pathgenic variant nor a benign variant, to our knowledge. The p.Arg320His variant has been reported with allele frequency of 0.0008% in gnomAD Exomes database and is novel (not in any individuals) in 1000 Genomes. This variant has not been reported to the ClinVar database. The amino acid Arg at position 320 is changed to a His changing protein sequence and it might alter its composition and physicochemical properties. For these reasons, this variant has been classified as a Variant of Uncertain Significance (VUS).
