NM_025114.4(CEP290):c.255T>A (p.Asn85Lys) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the CEP290 gene demonstrated a sequence change, c.255T>A, in exon 5 that results in an amino acid change, p.Asn85Lys. The p.Asn85Lys change affects a moderately conserved amino acid residue located in a domain of the CEP290 protein that is not known to be functional. The p.Asn85Lys substitution appears to be benign using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). This sequence change does not appear to have been previously described in individuals with CEP290-related disorders. Allele frequency data from population databases such as gnomAD may not be reliable for this sequence change. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Asn85Lys change remains unknown at this time.
