Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007294.4(BRCA1):c.202dup (p.Ile68fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 202, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 68, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ile68Asnfs*13) in the BRCA1 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). This variant has been reported in individuals with a personal and/or family history of breast and/or ovarian cancer (PMID: 29446198). ClinVar contains an entry for this variant (Variation ID: 266210). This variant is not present in population databases (ExAC no frequency).
