NM_000051.4(ATM):c.8564del (p.Ser2855fs) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 8564, deleting one base; at the protein level this means shifts the reading frame starting at serine residue 2855, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in ATM is denoted c.8564delG at the cDNA level and p.Ser2855MetfsX2 (S2855MfsX2) at the protein level. The normal sequence, with the base that is deleted in braces, is CGCA[G]TGTA. The deletion causes a frameshift which changes a Serine to a Methionine at codon 2855, and creates a premature stop codon at position 2 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. Based on the currently available information, we consider this deletion to be a likely pathogenic variant.
