This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 1650, deleting one base; at the protein level this means shifts the reading frame starting at lysine residue 550, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in PALB2 is denoted c.1650delA at the cDNA level and p.Lys550AsnfsX11 (K550NfsX11) at the protein level. The normal sequence, with the base that is deleted in braces, is ACAA[A]TATC. The deletion causes a frameshift which changes a Lysine to an Asparagine at codon 550, and creates a premature stop codon at position 11 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay.
