Pathogenic — the classification assigned by GeneDx to NM_001174147.2(LMX1B):c.227G>A (p.Trp76Ter), citing GeneDx Variant Classification (06012015): The W76X pathogenic variant in the LMX1B gene has been reported previously in association with nail-patella syndrome (Dunston et al., 2004). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The W76X variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret W76X as a pathogenic variant.
