Pathogenic — the classification assigned by GeneDx to NM_006846.4(SPINK5):c.136C>T (p.Gln46Ter), citing GeneDx Variant Classification (06012015): The Q46X pathogenic variant in the SPINK5 gene has been reported previously in association withNetherton syndrome, in an affected individual who was heterzygous for the Q46X variant and anotherpathogenic variant (Sprecher et al., 2001). This variant is predicted to cause loss of normal proteinfunction either through protein truncation or nonsense-mediated mRNA decay. The Q46X variant wasnot observed in approximately 5,900 individuals of European and African American ancestry in theNHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations.We interpret Q46X as a pathogenic variant.
