This variant was reported in individual(s) with features consistent with APC-related familial adenomatous polyposis (Lagarde A et al. J Med Genet, 2010 Oct;47:721-2; Cao X et al. Eur J Hum Genet, 2000 Jan;8:42-8; Ambry internal data). This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
