Uncertain significance — the classification assigned by GeneDx to NM_002055.5(GFAP):c.1003G>A (p.Gly335Arg), citing GeneDx Variant Classification (06012015): The G335R variant in the GFAP gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G335R variant was not observed at any significant frequency in approximately 6500 individuals of European and African American ancestry by the NHLBI Exome Sequencing Project. The G335R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Glycine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret G335R as a variant of uncertain significance
