This variant lies in the ABCA12 gene (transcript NM_173076.3) at coding-DNA position 3256, where A is replaced by T; at the protein level this means converts the codon for lysine at residue 1086 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The K1086X nonsense variant in the ABCA12 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Although the K1086X variant has not been published previously to our knowledge, we consider this variant to be pathogenic.
