Uncertain significance for Aortic valve disease 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005585.5(SMAD6):c.806_817+19del, citing Invitae Variant Classification Sherloc (09022015): This variant results in the deletion of part of exon 1 (c.806_817+19del) of the SMAD6 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SMAD6 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with SMAD6-related conditions. ClinVar contains an entry for this variant (Variation ID: 2645478). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
