Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.4316C>T (p.Ala1439Val), citing Ambry Variant Classification Scheme 2023. This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 4316, where C is replaced by T; at the protein level this means replaces alanine at residue 1439 with valine — a missense variant. Submitter rationale: The p.A1439V variant (also known as c.4316C>T), located in coding exon 34 of the FBN1 gene, results from a C to T substitution at nucleotide position 4316. The alanine at codon 1439 is replaced by valine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6494 samples (12988 alleles) with coverage at this position. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
