NM_001148.6(ANK2):c.10310T>C (p.Ile3437Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ANK2 gene (transcript NM_001148.6) at coding-DNA position 10310, where T is replaced by C; at the protein level this means replaces isoleucine at residue 3437 with threonine — a missense variant. Submitter rationale: The p.I3437T variant (also known as c.10310T>C), located in coding exon 38 of the ANK2 gene, results from a T to C substitution at nucleotide position 10310. The isoleucine at codon 3437 is replaced by threonine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6502 samples (13004 alleles) with coverage at this position. This amino acid position is poorly conserved in available vertebrate species; and threonine is the reference amino acid in three species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
