NM_017617.5(NOTCH1):c.7150C>G (p.Gln2384Glu) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2384E variant (also known as c.7150C>G), located in coding exon 34 of the NOTCH1 gene, results from a C to G substitution at nucleotide position 7150. The glutamine at codon 2384 is replaced by glutamic acid, an amino acid with highly similar properties. This variant has been detected in a sporadic bicuspid aortic valve cohort; however, details were limited (Debiec RM et al. Heart. 2022 Jun;108(14):1114-1120). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
