NM_000090.4(COL3A1):c.1813C>G (p.Gln605Glu) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the COL3A1 gene (transcript NM_000090.4) at coding-DNA position 1813, where C is replaced by G; at the protein level this means replaces glutamine at residue 605 with glutamic acid — a missense variant. Submitter rationale: The p.Q605E variant (also known as c.1813C>G), located in coding exon 25 of the COL3A1 gene, results from a C to G substitution at nucleotide position 1813. The glutamine at codon 605 is replaced by glutamic acid, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
