This variant lies in the FBN2 gene (transcript NM_001999.4) at coding-DNA position 6764, where A is replaced by G; at the protein level this means replaces asparagine at residue 2255 with serine — a missense variant. Submitter rationale: The p.N2255S variant (also known as c.6764A>G), located in coding exon 54 of the FBN2 gene, results from an A to G substitution at nucleotide position 6764. The asparagine at codon 2255 is replaced by serine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
