Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.11234G>A (p.Gly3745Glu), citing Ambry Variant Classification Scheme 2023: The p.G3745E variant (also known as c.11234G>A), located in coding exon 46 of the AKAP9 gene, results from a G to A substitution at nucleotide position 11234. The glycine at codon 3745 is replaced by glutamic acid, an amino acid with similar properties. This alteration has been reported in a control population in a cancer predisposition case control study (Pritchard AL et al. PLoS One, 2018 Apr;13:e0194098). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
