This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 149, where A is replaced by T; at the protein level this means replaces histidine at residue 50 with leucine — a missense variant. Submitter rationale: The p.H50L variant (also known as c.149A>T), located in coding exon 1 of the FBN1 gene, results from an A to T substitution at nucleotide position 149. The histidine at codon 50 is replaced by leucine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6493 samples (12986 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species.
