Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.5140G>A (p.Ala1714Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the FLNA gene (transcript NM_001110556.2) at coding-DNA position 5140, where G is replaced by A; at the protein level this means replaces alanine at residue 1714 with threonine — a missense variant. Submitter rationale: The p.A1706T variant (also known as c.5116G>A), located in coding exon 29 of the FLNA gene, results from a G to A substitution at nucleotide position 5116. The alanine at codon 1706 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6401 samples with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
