This variant lies in the RAF1 gene (transcript NM_002880.4) at coding-DNA position 142, where C is replaced by G; at the protein level this means replaces leucine at residue 48 with valine — a missense variant. Submitter rationale: The p.L48V variant (also known as c.142C>G), located in coding exon 1 of the RAF1 gene, results from a C to G substitution at nucleotide position 142. The leucine at codon 48 is replaced by valine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in mammals but not in all available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
