Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_030777.4(SLC2A10):c.119T>C (p.Phe40Ser), citing Ambry Variant Classification Scheme 2023: The p.F40S variant (also known as c.119T>C), located in coding exon 2 of the SLC2A10 gene, results from a T to C substitution at nucleotide position 119. The phenylalanine at codon 40 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
