Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.698A>T (p.Gln233Leu), citing Ambry Variant Classification Scheme 2023: The p.Q233L variant (also known as c.698A>T), located in coding exon 4 of the TGFB3 gene, results from an A to T substitution at nucleotide position 698. The glutamine at codon 233 is replaced by leucine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6,503 samples (13,006 alleles) with coverage at this position. This amino acid position is conserved in available vertebrate species through reptiles. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
